Cathedral

I like to ask about this project's specialisation.

It has a total of 4.
1) God of the Hunt +10 Food and +5 Wealth for each Animal within borders. Stacks up to 3 times
2) Relics - The Cathedral has 2 mystic 'items' as specialisation that give +5 tech if turned into Relics. +100 food +100 wealth +100awe if all items are Relics
3) Shell Piece - +15 food for each animal transmuted at most once within borders. Become a relic if activated at least 3 times
4) Evermuck - +15 wealth for each animal with at least 2 Danger within borders. Become a relic if activated at least 3 times

I manage to activate both 3 and 4 but I did not see 2 being activated.

Any idea why?

Did you activate 3) and 4) three times? (eg 3 animals transmuted at most once, 3 animals with 2+ danger).
That's when those become relics and activate 2).

Yap I did.
As I have activated both 3) and 4), as in 3 animals transmuted at most once and 3 animals with 2+ danger
I got the bonus added under 3) (+75 food) 4) (+15 wealth) but 2) not given at all.
